ASCO publishes Living Guidelines for MM treatment
The American Society of Clinical Oncology (ASCO) published Living Guidelines outlining recommendations for multiple myeloma (MM) treatment. The ASCO guidelines were published online ahead of the print issue of the Journal of Clinical Oncology on May 20, 2026.
First dose of in vivo MM CAR-T therapy administered in the United States
The phase 1 MMyCAR trial of KLN-1010, an in vivo BCMA-targeted CAR-T therapy, currently ongoing in Australia, has expanded to the United States; the first US patient with relapsed/refractory MM (RRMM) received KLN-1010, according to a news release. Preliminary efficacy data from MMyCAR were presented at the Late-Breaking Abstracts Session at the 2025 American Society of Hematology Annual Meeting. In vivo approaches for T-cell engaging therapies were explored in the closing session of the 7th Immune Effector Cell Therapies in Multiple Myeloma Workshop, held March 27–28, 2026.
Real-world evidence supports efficacy of cilta-cel in early relapsed myeloma
The BCMA-directed CAR-T therapy ciltacabtagene autoleucel (cilta-cel) demonstrated deep responses and a 12-month progression-free survival (PFS) rate of 79% in patients with RRMM who received the treatment after 1–3 prior lines, consistent with results of the CARTITUDE-4 trial. These findings from a real-world cohort in the German Registry for Stem Cell Transplantation and Cellular Therapy were reported in the Journal of Hematology and Oncology.
MM research presented at ASCO 2026 includes AI tools, clinical trial data
Myeloma research presented at the 2026 Annual Meeting of the American Society of Clinical Oncology included data from clinical trials, an AI-enabled tool for predicting therapeutic benefit based on routine bone marrow biopsy imaging slides, and a stepwise prompting approach to facilitate ChatGPT-aided MM diagnosis.
In the MajesTEC-9 study, teclistamab improved PFS in patients with RRMM who had not received prior BCMA-directed therapy (BCMA-naïve), compared with investigator’s choice of pomalidomide- or carfilzomib-based triplets.

Exploratory analysis compares cost implications of bispecific antibody therapies
In a study reported in Current Medical Research and Opinion, researchers used a matching-adjusted indirect comparison model to assess the cost implications for treatment with different bispecific antibodies in triple drug-class-exposed RRMM. Total costs for linvoseltamab were lower than teclistamab at year one and two and comparable to elranatamab, per the model assumptions and analyses.
Elranatamab demonstrates durable responses in heavily pretreated patients
In a subgroup of BCMA-naïve patients with RRMM enrolled in the MagnetisMM-3 phase trial, elranatamab demonstrated an overall response rate of 66%. The post hoc analysis, published in Cancer, showed a median duration of response of 40.8 months, with median PFS and OS values of 27.3 and 43.6 months, respectively.
