Plasma exchange may aid kidney recovery in newly diagnosed patients with light chain cast nephropathy
While plasma exchange and daratumumab are often used to treat light chain cast nephropathy in patients with multiple myeloma (MM), neither approach was associated with kidney recovery at 90 days, according to a cohort study conducted across 25 US academic centers. There was, however, an association between plasma exchange and kidney recovery at 90 days in patients who were newly diagnosed, but not in those with relapsed/refractory MM. These findings were published in the American Journal of Hematology.
Isatuximab quadruplet yields sustained MRD negativity in high-risk transplant-ineligible NDMM
In a longitudinal analysis of the phase 3 BENEFIT trial, isatuximab combined with bortezomib-lenalidomide-
Long-term MAIA data support benefit of daratumumab triplet in frail patients
A retrospective subgroup analysis of the phase 3 MAIA trial showed that daratumumab, combined with Rd, improved progression-free survival as well as the overall rates and depth of response in transplant-ineligible patients with NDMM, compared with Rd alone, regardless of frailty status at baseline. These findings were reported in Leukemia.
Dual-antigen targeting may help avoid antigen escape-mediated resistance
According to a report in Hemasphere, the BCMA×GPRC5D dual-targeted trispecific antibody ramantamig demonstrated higher potency in inducing MM cell lysis than a combination of two different bispecific antibodies targeting either BCMA or GPRC5D. Ramantamig’s activity was also less sensitive to the presence of soluble BCMA.
Chinese MM guidelines integrate IMS-IMWG CGS criteria
The 2026 revision of the guidelines for MM diagnosis and treatment from the Chinese Medical Doctor Association, Chinese Society of Hematology, and Chinese Medical Association, published in Zhonghua Nei Ke Za Zhi, includes both the Chinese comprehensive risk-stratification system and the 2025 IMS-IMWG Consensus Genomic Staging criteria for high-risk MM.
In vivo BCMA CAR T-cells have limited persistence despite early efficacy signals
In vivo BCMA-directed CAR-T therapy, which yielded 100% objective response rate at up to 3 months of follow-up, demonstrated limited persistence in three out of four patients over an extended follow-up, up to 15 months. While three patients experienced relapse or progression, the longer-term data reported in Nature Medicine showed robust CAR-T expansion and durable response in one patient. “Further optimization of the in vivo CAR-T platform” is needed, authors concluded.
The International Myeloma Society is a professional, scientific, and medical society established to bring together clinical and experimental scientists involved in the study of myeloma. The purpose of this society is to promote research, education, clinical studies (including diagnosis and treatment), workshops, conferences, and symposia on all aspects of multiple myeloma worldwide.
The IMS is a membership organization comprised of basic research scientists, and clinical investigators in the field along with physicians and other healthcare practitioners.
